Laboratory autoclave

2023-03-22
Laboratory autoclave (also called autoclave, autoclave or pressure cooker) is a sterilization device widely used in biology, chemistry, medicine, food and other fields, especially for killing bacteria, viruses, fungi and other microorganisms. It sterilizes under high pressure through steam to ensure that the instruments, culture media, solutions, equipment, etc. used in the experiment or production process meet the sterility standards.

Working Principle
The basic principle of high-pressure steam sterilizer is to use the high temperature and high pressure of saturated steam to make the surface and internal temperature of the sterilized items reach a certain standard under heating and pressurization, so as to kill microorganisms. Its core processes include:
Heating and pressurization: Introducing high-temperature steam into the sterilizer through a steam generator or steam pipe to increase the temperature and pressure.
The role of temperature and pressure: High-pressure steam can quickly heat the sterilized items to a high temperature range of usually 121°C to 134°C, thereby effectively destroying the cell walls, proteins and nucleic acids of bacteria and killing microorganisms.
Insulation time: In order to ensure thorough sterilization, the sterilizer needs to be kept at a specific temperature for a period of time, usually between 15-30 minutes, depending on the temperature and the type of item.
Cooling and releasing pressure: After sterilization is completed, the equipment will decompress, allowing the cooling process to occur, and finally release steam to remove the sterilized items.

Common types

Vertical high pressure steam sterilizer
Features: Traditional sterilizer, the sterilization chamber is set vertically, and the appearance is similar to a large boiler. Suitable for small and medium-sized laboratories.
Application: Commonly used for sterilization of medical equipment, glassware, culture medium and other items.
Advantages: Simple structure, easy operation, suitable for processing small amounts of samples.

Horizontal high pressure steam sterilizer
Features: The sterilization chamber is placed horizontally and has a large sterilization capacity. It can handle larger batches of items.
Application: Suitable for large-scale laboratories or production lines, especially in the medicine, pharmaceutical and food industries.
Advantages: Large capacity, suitable for batch processing, and more uniform during the sterilization process.

Vertical intelligent high-pressure steam sterilizer
Features: Equipped with advanced automatic control system, it can accurately control parameters such as temperature, time, pressure, etc., and automatically complete the sterilization process.
Application: Suitable for laboratories and biological research fields that require high-precision sterilization.

Advantages: Simple operation, programmed control, can be operated through the touch screen, with self-test and self-diagnosis functions, providing higher safety and reliability.

Desktop high pressure steam sterilizer
Features: Small structure, suitable for laboratory use, especially for small laboratories with limited space.
Application: Used for small batch instrument sterilization, culture medium sterilization, etc.
Advantages: Small footprint, suitable for small laboratories, easy to operate.

Main components
Sterilization chamber
This is the core part of the autoclave, which is used to place the items to be sterilized. It is usually made of stainless steel with a smooth inner wall for easy cleaning and maintenance.
Heating system
Steam is provided by electric heating or steam heating system, usually by heating coils or boilers.
Pressure control system
The internal pressure is regulated and monitored by equipment such as safety valves and pressure gauges to ensure that the appropriate pressure is maintained during the sterilization process.
Temperature control system
Temperature sensors and temperature control devices are used to maintain the set temperature to ensure the sterilization effect. Most modern autoclaves are also equipped with digital displays that can display temperature changes in real time.
Exhaust system
It is used to release the high-pressure steam in the chamber after sterilization is completed. It is usually equipped with an automatic exhaust valve to ensure smooth pressure release and avoid any safety problems.
Automatic control system
PLC control system, touch screen operation interface and programmed control, etc. Modern autoclaves are usually equipped with intelligent automatic control systems that can accurately control parameters such as sterilization temperature, time and pressure to avoid human operation errors.
Safety devices
Include safety valves, temperature protection devices, automatic power off systems, etc. to ensure safe operation during sterilization.

Usage process
Preparation:
Wash the items to be sterilized and put them into the sterilizer, ensuring that the items can flow steam freely.
Set the sterilizer program and select the appropriate temperature and time.
Heating and pressurizing:
Start the equipment, the heater starts to heat, and the steam gradually fills the sterilization chamber. As the temperature rises, the internal pressure gradually increases.
Sterilization process:
Maintain a certain time at the set temperature and pressure to ensure that the microorganisms are completely sterilized. The common standard is 121°C, 15-20 minutes, but different items may require different temperatures and times.
Cooling and exhaust:
After the sterilization is completed, the equipment begins to cool automatically, and the decompression system releases steam to cool down to a safe temperature.
Remove items:
Wait for the pressure of the equipment to be completely released, open the sterilizer, and take out the sterilized items. At this time, the items are in a sterile state and can be operated subsequently.
Common applications
Laboratory sterilization: used for sterilization of culture media, reagents, and experimental equipment (such as glassware, metal tools, etc.). Laboratory high-pressure steam sterilizers are often used in experiments such as biology, microbiology, and chemical analysis to ensure the sterility of the experimental environment.
Medical device sterilization: In medical places such as hospitals and clinics, sterilizers are used to sterilize disposable items such as instruments, syringes, and needles to prevent cross infection.
Food and beverage industry: Some equipment, containers or raw materials in the production process need to be sterilized to prevent microbial contamination.
Pharmaceutical industry: Used to sterilize drugs, medicinal instruments, etc. to ensure the sterile conditions of the pharmaceutical process.
Advantages
Efficient sterilization: It can effectively kill microorganisms such as bacteria, viruses, and fungi, and is widely used in aseptic processing.
Accurate temperature control: It can accurately control temperature and pressure to ensure the stability and repeatability of sterilization.
Wide applicability: It is suitable for sterilization of different types of experimental materials, medical devices, foods, and drugs.
High safety: Modern high-pressure steam sterilizers are equipped with multiple safety devices, such as pressure safety valves, temperature control, and automatic exhaust systems, which effectively avoid dangers during operation.

Precautions
Pre-operation inspection: Ensure that the equipment is fault-free and the pressure gauge and temperature control system are working properly.
Regular maintenance: Clean the sterilizer regularly, check the sealing ring, heating system, and safety valve, etc., to ensure long-term and stable operation of the equipment.
Item placement: When placing items, be careful not to over-stack them to avoid affecting steam circulation.
Laboratory high-pressure steam sterilizers are common and essential equipment in laboratories and production environments. They can effectively kill microorganisms through high-temperature and high-pressure steam to ensure the safety and sterility of operations. With the continuous development of technology, modern high-pressure steam sterilizers are gradually developing in the direction of automation and intelligence, further improving the safety, efficiency and accuracy of operations.
